Although the exact causes of psoriasis remain unknown, genetic factors are believed to play a critical role in its development. Several Genome-Wide Association Studies (GWAS) have identified multiple susceptibility loci associated with psoriasis, indicating that multiple genes are involved in its pathogenesis.Objectives:The objective of my study is to clone the susceptibility genes of psoriasis and construct a transgenic mouse model to investigate their roles in the disease pathogenesis.Methods:1. Cloning of susceptibility genes: The candidate susceptibility genes will be identified from public databases, such as GWAS and expression profile analysis. The genes will be cloned using polymerase chain reaction (PCR) and subsequently validated using DNA sequencing.2. Construction of transgenic mouse model: The cloned genes will be inserted into a plasmid vector containing a tissue-specific promoter, such as the K14 promoter, which drives gene expression specifically in the epidermis. The plasmid will then be microinjected into fertilized oocytes of mice. Founders with germline transmission of the transgene will be identified by PCR and confirmed by Southern blotting.3. Characterization of transgenic mice: The phenotype of transgenic mice will be assessed by histology, immunohistochemistry, real-time PCR, and Western blot analysis. The findings of this project will have important implications for the development of new therapies for psoriasis.
