精品文档---下载后可任意编辑一株产大环内酯类抗真菌抗生素菌株的鉴定及发酵工艺优化的开题报告摘要:本文旨在鉴定并优化一株产大环内酯类抗真菌抗生素的菌株的发酵工艺。通过初步筛选,从土样中分离出具有抗真菌活性的菌株,经鉴定为链霉菌属菌株。基于菌株的生长特性,通过单因素实验和正交实验优化了发酵工艺,并利用高效液相色谱对发酵产物进行了分析。优化后的发酵条件为:初始 pH 值 7.0、温度 30 ℃、初始接种量 3%,添加玉米粉 3%、添加发酵辅助剂 MgSO4·7H2O 0.05%、CuSO4·5H2O 0.005%,培育 48h。分离出的抗真菌活性物质经 HPLC 分析得出主要产品结构为大环内酯类抗生素。关键词:大环内酯;抗真菌;链霉菌;鉴定;发酵工艺Abstract:This paper aims to identify and optimize the fermentation process of a strain of macrocyclic lactone antifungal antibiotic-producing bacteria. Through preliminary screening, a strain with antifungal activity was isolated from soil samples and identified as a Streptomyces species. Based on the growth characteristics of the strain, the fermentation process was optimized through single-factor experiments and orthogonal experiments, and the fermentation products were analyzed by high-performance liquid chromatography. The optimized fermentation conditions were: initial pH value of 7.0, temperature of 30 ℃, initial inoculation amount of 3%, addition of 3% corn starch, addition of fermentation adjuvants MgSO4·7H2O 0.05% and CuSO4·5H2O 0.005%, and incubation for 48h. The isolated antifungal active substance was analyzed by HPLC, and the main product structure was identified as macrocyclic lactone antibiotic.Keywords: macrocyclic lactone; antifungal; Streptomyces; identification; fermentation process
